It, therefore, poietic cells. appears that human hemopoietic cells (like I T HAS BEEN APPRECIATED for some time that folate occurs in natural materials largely with three or more glutamic acid residues attached to the pteroyl portion of the molecule. During absorption of food folates, these folate polyglutamates are enzymatically hydrolyzed to the equivalent monoglutamate derivatives.’ It is still uncertain where this hydrolysis occurs, but the finding that the enzyme involved, y-glutamyl carboxypeptidase (folate conjugase, pteroylpolyglutamate hydrolase), is found within the lysosomes of the small intestinal cells2 raises the possibility that conjugated folates may enter the intestinal cell as such before hydrolysis to the monoglutamate form.3 Some bacterial cells (e.g., L. casei) are capable of transporting triglutamate forms of folate, while others (e.g., S. fecalis) cannot transport compounds with more than two glutamate moieties.4 No equivalent information is available, however, about transport of conjugated folates into animal or human cells. The demonstration that polyglutamyl analogues of at least some antifolates are more effective in vitro than the corresponding monoglutamate forms5 and the suggestion that antifolate polyglutamates might be used in chemotherapy#{176} make the question of the cellular transport of conjugated folates more urgent. Previous work has shown that dU (deoxyuridine) does not inhibit the uptake

On-demand cellular uptake of cysteine conjugated gadolinium based mesoporous silica nanoparticle with breast cancer-cells

Design, synthesis, and conjugation of mesoporous silica nanoparticles (MSNs) with biomolecules is a matter of growing interest to enhance selective uptake of contrast agents like gadolinium (Gd3+) by cancer cells. Here, by targeting xc-cystine/glutamate antiporter system in breast cancer cells, conjugation of MSN-Gd3+ with cysteine is used to enhance cancer cellular uptake of Gd3+. Reactions de...

Synergistic cellular toxicity and uptake effects of iodixanol conjugated to anionic linear globular dendrimer G2

Objective(s): Early diagnosis of cancer using noninvasive imaging techniques has been discussed in several recent studies. The present study aimed to assess the synergistic effects of iodixanol-conjugated polyethylene glycol (PEG)-citrate (anionic linear globular) dendrimer G2 on MCF-7 breast cancer cells and human embryonic kidney 293 (HEK293) cells. Materials and Methods: PEG-citrate de...

The effect of folate analogues on the thymidine utilization by human and rat marrow cells and the effect on the deoxyuridine suppression test.

The deoxyuridine (dU) suppression test assesses the capacity of marrow cells or activated lymphocytes to convert exogenous dU into thymidine. In addition, the effect of the added cobalamin or folate on the result is used to determine the nature of the deficiency in megaloblastic marrows. This study shows that folates have a considerable effect on the uptake of labelled thymidine by marrows from...

Cytotoxicity of 5-ALA-conjugated bismuth oxidenanoparticles on KB cell line

Introduction: In recent years, bismuth-based nanomaterials have been widely used in medical researches as imaging, drug delivery and x-ray radiosensitizing agents. Due to their anti-microbial effects against Helicobacter pylori (HP), bismuth colloidal compounds are used to treat various types of diseases such as chronic gastritis. Despite their advantages, bismuth-based compoun...

Comparative Analysis of Mesenchymal Stem Cells Isolated from Human Bone Marrow and Wharton’s Jelly

Introduction: Bone marrow (BM) is a known source of mesenchymal stem cells (MSCs) that are used for cell therapy. This study attempts to identify if the Wharton’s Jelly (WJ) is a suitable substitute for BM as a source for MSCs. Materials and Methods: A population of human WJ and BM stem cells were isolated and incubated with fluorescein conjugated antibodies for five specific MSC markers....
